Fibroblasts

Fibroblasts are cells that synthesize and maintain the extracellular matrix of many tissues. Fibroblasts provide a structural framework (stroma) and play a critical role in processes such as wound healing. Fibroblasts are the most common cells of connective tissues. Fibroblasts continuously secrete pre-cursors of the extracellular matrix. The cells are morphologically heterogeneous. Their appearance is depending on location and cellular activity.  Fibroblasts do not form flat monolayers and are not restricted by a polarizing attachment to basal lamina. Fibroblasts can also slowly migrate over substratum as individual cells.  Fibroblasts are derived from mesenchyme. They express vimentin, a marker often used to identify their mesodermal origin. Fibroblasts have a branched cytoplasm surrounding elliptical, speckled nucleus having 1 or 2 nucleoli. Active fibroblasts are characterized by abundant rough endoplasmic reticulum. Inactive fibroblasts (referred to as fibrocytes) are smaller and spindle shaped. Fibrocytes have significantly reduced endoplasmic reticulum. Fibroblasts, when confluent, often align parallel in clusters. Fibroblasts generate collagens, glyosaminoglycans, reticular and elastic fiber, and glycoproteins identified in the extracellular matrix. Tissue damage stimulates fibrocytes and induced mitosis of fibroblasts to divide and synthesize ground substance.  Fibroblast can give rise to other cells, such as bone cells, fat cells and smooth muscle cells. All these lineages are of mesodermal origin. Fibroblasts have a high proliferation rate and therefore are a popular cell type for research.
