CellMade Human Disease Primary Liver Fibroblasts (HLiF) and Pancreatic Fibroblasts (HPaF)

Product Description

  • HLiF are isolated from normal human liver tissue.
  • HPaF are isolated from normal human pancreatic tissue.
  • Human tissues are sourced via CellMade’s own tissue acquisition network under standardized conditions (temperature, transport medium, transport, delay, …). CellMade manages its own biobank and guarantees full compliance and traceability according to the laws of France.

Isolation method

The method we use to isolate primary fibroblasts was developed based on a combination of established and proprietary methods.  Fibroblasts are pre-coated with fibroblast-specific antigen antibody, and selected with magnetic beads pre-coated with secondary antibody.

Fibroblasts are grown in T75 tissue culture flasks pre-coated with gelatin-based coating solution for t = 2 min and incubated in CellMade’s  Complete Growth Medium, depending on the selected primary cell model, generally for 3-7 days. Cultures are then expanded. Cells at passage 1 are detached from cell culture flasks and immediately cryopreserved. Each vial contains at least 0.5x106 cells per mL.

Cells can be expanded for up to five (5) passages under the cell culture conditions specified by CellMade.

Product Testing and Functional Characterization

  • HLiF and HPaF from CellMade display typical fusiform appearance under light microscopy.
  • Standard: HLiF and HPaF are tested negative for the expression of the von Willebrand Factor VIII, cytokeratin 18, and α-smooth muscle actin by immunofluorescence staining methods and cytometry.
  • Functionally characterized: Transforming growth factor-ß (TGF ß) stimulation of total protein and collagen synthesis, measured in time-response/dose -response manner via total protein measurement and ELISA.
  • All CellMade primary cell models test negative for TVO, Moulds, Yeasts and Mycoplasmas as tested by PCR methods.
  • All CellMade primary cell models test negative for HIV-1, HIV-2, Hepatitis-B and Hepatitis-C as tested by PCR methods and Antibody-methods.
  • All CellMade primary cell models are accompanied by a Certificate of Analysis.

Biochemical characterization

  • CellMade has established rigorous biochemical quality control programs ensuring the qualification of normal and disease primary cell models.
