GMO Screening

CellMadeTM has chosen to implement SphaeraMag® DNA extraction and PhoenixDx® PCR methodologies developed by Procomcure Biotech (Salzburg, Austria). Procomcure Biotech provides exceptional validation of their extraction and PCR kits, which sets them apart from other food allergen PCR kit suppliers.

The first step in the analysis process to detect, identify and quantify GMO events in feed, food and other products is screening. In a typical screening approach, an unknown sample is analyzed by a first PCR Array detecting species specific DNA. The results enable to identify the presence of e.g., maize, rapeseed, or soybean. As an example: in case this analysis renders a positive result for maize and negative results for rapeseed and soybean, then a second PCR array is used for the detection of all EU authorized maize GMO events. In case this analysis renders a positive for two (or more) species, then several PCR arrays are used.

The sensitivity and reliability of Real-time PCR Arrays are related to the quality and performance of the DNA extraction protocol. DNA damage during extraction will lead to reduced efficiency in the PCR Array reaction. Samples containing high levels of carbohydrates, proteins and lipids may inhibit the PCR reaction. To increase efficacy, proprietary DNA extraction protocols were developed in cooperation with Procomcure Biotech (Salzburg, Austria). These protocols remove polysaccharides (like pectin, cellulose, hemi-cellulose, starch, etc.) and proteins. DNA is reversibly bound to silica-coated magnetic beads and the final elution step renders highly pure and concentrated DNA. The DNA extraction method is automated.

The DNA extraction methods and Real-time PCR Arrays have been validated according to the principles and requirements of international standards and assure consistent and reproducible results.

Detailed reports are provided summarizing test findings for in-process and batch release testing, along with insights for validation purposes. CellMadeTM Laboratories employs PCR methods selected from trusted suppliers and participates in international laboratory proficiency tests to ensure accuracy.
